Hey, you spotted the scan lines weren’t folds so kudos there. It probably didn’t get much attention because it wasn’t arguable lol. We do like a good arguem….I mean discussion.

It is basically a redistribution of the gray-scales and blacks of the letters so that the darker areas (more inked) are more pronounced. Darker areas are typically where strokes connect and this gives insight on how Zodiac wrote the symbols in the 340. An example is the following, the squares always start in the upper left corner. As far as I can tell all symbols have been written in the same manner and I feel that this level of consistency may be unusual. Usually people deviate here and there, but not Zodiac.
Triangles start in the lower left corner.

Here are the line symbols. Horizontal lines drawn from right-to-left and vertical and diagonal lines from bottom-to-top. The 3rd horizontal line symbol may be an exception.

No. It requires a uniformly lit high resolution scan of a message written preferably with some kind of felt-tip pen.
Circle shaped symbols start and close in the upper center.
Plus symbols are heavier/thicker/darker on the right and/or bottom sides, this is consistent with the way the line symbols are drawn as seen a couple of posts back. I marked only a few examples just to give the idea of where to look. Of course you can also see darkening in the middle of the symbols as this is where the lines crossed.

this is really a very interesting investigation. I think it’s very important to know how z340 was made. However, I must disagree with you in your conclusion. I think the strokes were drawn the other way around. Take a felt pen and try it out on a piece of paper. Especially if it is a very wet felt pen, a small drop forms as soon as you have finished a line and lift it up again. Here is an example (strokes drawn from left to right):
